Design and in vitro characterization of PAC1/VPAC1-selective agonists with potent neuroprotective effects 阐明了PACAP氨基末端的分子结构特征,并开发能促进神经元存活的强效选择性PAC1/VPAC1受体激动剂。并合成了一系列PACAP衍生物,分别检测了它们对PAC1、VPAC1和VPAC2受体的结合亲和力及诱导细胞内钙动员的能力。最终,在神经元损伤体外模型中评估了具有优化药理学特性的类似物。
研究结果显示:在第2位引入羟脯氨酸或第7位引入丙氨酸的衍生物均未表现出显著VPAC2受体激动活性。构效关系研究表明,可能存在共同(Asx-turn样)和独特(不同N端封盖类型)的二级结构,这些结构可能决定受体的识别、选择性和激活。值得注意的是,在预处理和共处理实验中,[Ala7]PACAP27和[Hyp2]PACAP27均展现出显著保护人多巴胺能SH-SY5Y神经母细胞免受MPP+毒性的能力。这些新的药理学和结构数据将为合理设计PACAP衍生化合物提供重要依据,这类化合物有望成为治疗神经退行性疾病的潜在治疗药物。
体外研究
PACAP (1-38),human,ovine,rat 是垂体腺苷酸环化酶激活多肽的片段。
PACAP (1-38) 对来自各种组织 (包括内分泌胰腺) 的膜中的 PACAP 特异性 (PAC1) 受体表现出高亲和力。
在体外,PACAP (1-38) 松弛由组胺和乙酰胆碱预收缩的豚鼠和兔气管平滑肌。PACAP (1-38) 还增加气管平滑肌中的腺苷 3':5'-环磷酸 (环 AMP),为 PACAP (1-38) 的松弛作用提供了可能的机制。
体内研究
在假动物中单独使用 PACAP (1-38) 不会导致任何视网膜层发生变化。PACAP (1-38) 溶解在 solutio ophthalmica cum benzalkonio 中,可显著保护双侧颈总动脉闭塞 (BCCAO) 受损视网膜的视网膜;与对照视网膜相比,用 PACAP (1-38) 滴眼液处理的视网膜结构得以保留。与假手术对照组相比,BCCAO 视网膜中的 OLM-ILM (外界膜-内界膜) 距离减少了 49.7% (p<0.001),但在接受 PACAP 处理的眼睛中仅为 40.6% (p<0.001) (1-38) 眼药水。在内核层 (INL) (BCCAO:38.5%,PACAP (1-38):30.5%;p<0.001) 和内丛状层 (IPL) (BCCAO:64.8%,PACAP (1-38):38.2%;p<0.05),而在外核层 (ONL) 中未观察到具有统计学意义的损伤衰减 (BCCAO:36.5%,PACAP (1-38):37.7%) 或外丛状层 (OPL) (BCCAO:53.0%,PACAP (1-38):48.2%)。BCCAO 后神经节细胞层 (GCL) 细胞数量显著减少 52.4% (p<0.05),PACAP (1-38) 滴眼液显著改善 (减少 25.9%;p<0.05)。

多肽H2N-His-Ser-Asp-Gly-Ile-Phe-Thr-Asp-Ser-Tyr-Ser-Arg-Tyr-Arg-Lys-Gln-Met-Ala-Val-Lys-Lys-Tyr-Leu-Ala-Ala-Val-Leu-Gly-Lys-Arg-Tyr-Lys-Gln-Arg-Val-Lys-Asn-Lys-NH2的合成步骤:
1、合成MBHA树脂:取若干克的MBHA树脂(如初始取代度为0.5mmol/g)和1倍树脂摩尔量的Fmoc-Linker-OH加入到反应器中,加入DMF,搅拌使氨基酸完全溶解。再加入树脂2倍量的DIEPA,搅拌混合均匀。再加入树脂0.95倍量的HBTU,搅拌混合均匀。反应3-4小时后,用DMF洗涤3次。用2倍树脂体积的10%乙酸酐/DMF 进行封端30分钟。然后再用DMF洗涤3次,甲醇洗涤2次,DCM洗涤2次,再用甲醇洗涤2次。真空干燥12小时以上,得到干燥的树脂{Fmoc-Linker-MHBA Resin},测定取代度。这里测得取代度为 0.3mmol/g。结构如下图:
2、脱Fmoc:取1.13g的上述树脂,用DCM或DMF溶胀20分钟。用DMF洗涤2遍。加3倍树脂体积的20%Pip/DMF溶液,鼓氮气30分钟,然后2倍树脂体积的DMF 洗涤5次。得到 H2N-Linker-MBHA Resin 。(此步骤脱除Fmoc基团,茚三酮检测为蓝色,Pip为哌啶)。结构图如下:
3、缩合:取1.02mmol Fmoc-Lys(Boc)-OH 氨基酸,加入到上述树脂里,加适当DMF溶解氨基酸,再依次加入2.03mmol DIPEA,0.97mmol HBTU。反应30分钟后,取小样洗涤,茚三酮检测为无色。用2倍树脂体积的DMF 洗涤3次树脂。(洗涤树脂,去掉残留溶剂,为下一步反应做准备)。得到Fmoc-Lys(Boc)-Linker-MBHA Resin。氨基酸:DIPEA:HBTU:树脂=3:6:2.85:1(摩尔比)。结构图如下:
